ABSTRACT

[Objective] To explore the methods of avoiding hemolysis in preparing serum complements of guinea pigs. [Methods] In method 1, the serum of 8 guinea pigs was fixed first and then sheep red blood cells (SRBC) were added to the mixed serum for absorption. In method 2, SRBC was added to the serum of 8 guinea pigs respectively for absorption first and then the serum containing SRBC was mixed. After the experiment, macroscopic examination was carried out to observe the hemolysis and the optical density of the two kinds of mixture was detected at the wavelength of 540 nm. [ Results] In the serum complements of guinea pigs prepared by method 1, macroscopic hemolysis occurred; in the serum complements prepared by method 2, macroscopic hemolysis was not obvious and the optical density of the mixture was lower than that by method 1 ( P

ABSTRACT

Different varieties of Terminalia chebula Retz. were identified by DNA finger prints, andat the same time, some problems encountered during the application of RAPD for the identification ofTCM were studied. The polymorphic features of DNA finger prints of different species were compared. Results of the study showed that they were differently related in heredity which can provide a basis for thedifferentiation of one species from the other. Means for the solution of problems arised during RAPD ap-plication were suggested. It was concluded that RAPD is an effective method to identify T. chebula from itsconfusable varieties.

ABSTRACT

【Objective】The effect of sinomenine on human cyclo-oxygenase(COX) and interleukin-1?(IL-1?) was observed to explore the anti-inflammatory mechanism of sinomenine.【Methods】Peripheral blood monocytes(PBMC) were isolated and incubated with sinomenine. After incubation, total RNA from PBMC was extracted and the effect of sinomenine on COX and IL-1? expression with or without the stimulation of lipopolysaccharide(LPS) was observed by the method of semi-quantitative reverse transcription polymerase chain reaction(RT-PCR).【Results】Sinomenine had no effect on COX-1 gene expression, had a slight inhibitory effect on COX-2 gene expression and had a strong inhibitory effect on IL-1? gene expression.【Conclusion】 The anti-inflammatory mechanism may be related to the inhibition of inflammatory cytokines.

ABSTRACT

Objective To study the effect of Dahuang Mudan Decoction on experimental colitis induced by trinitrobenzene sulfonic acid(TNBS)in mice and to explore its mechanism. Methods Mice colitis model was established by rectal administration of trinitrobenzene sulfonic acid.Disease activity index(DAI)and histological scores were assessed, the serum level of TNF-? was detected by ELISA.Results Dahuang Mudan Decoction reduced DAI and histological scores in mice, and alleviate inflammation in colonic tissue. It also significantly decreased mice serum TNF-? level. Conclusions Dahuang Mudan Decoction shows positive therapeutic efficacy on mice colitis induced by trinitrobenzene sulfonic acid, and its mechanism may be involved in inhibiting the production of TNF-?.

ABSTRACT

Objective To observe the effect of naringin on proliferation and differentiation of 3T3-L1 preadipocyte, and to investigate its possible mechanism. Methods The cultured 3T3-L1 cells were pretreated with naringin, and then MTT method was used to detect the proliferation of the cells, oil red O staining method and spectrophotography were applied to analyze the degree of differentiation. The expression of PPAR? 2 and C/EBP? mRNA associated with adipocytes differentiation were detected by reverse transcription PCR(RT-PCR).Results Naringin suppressed the proliferation and differentiation of 3T3-L1 preadipocytes and down-regulated the expression of PPAR? 2 and C/EBP? mRNA.Conclusion Naringin can inhibit the proliferation of 3T3-L1,and can suppress their differentiation through down-regulating the expression of PPAR? 2 and C/EBP? mRNA.

ABSTRACT

Objective To compare the genetic difference of Pogostemon cablin from different habitats. Methods Forty-six primers were used to analyze the fingerprint of Pogostemon cablin by Random Amplified Polymorphic DNA,and 6 of them were identifiable.Primer S358 could reflect the tiny polymorphic differences of Pogostemon cablin from different habitats which had 3 same amplified strips.Primers S358,S359,S443 could be used for the identification of Shipai sample from its other species. Results The results of cluster analysis showed that the two samples from Gaoyao were classified as one type, the four samples from Zhanjiang,Hainan,Shipai as another type, one sample from Caizhilin and one from the pharmacy of the First Affiliated Hospital of Guangzhou University of TCM as the the third type, and the sample from the Qingping Traditional Chinese Herbal Market as the fourth type. Conclusion The RAPD analysis can be used for the identification of Pogostemon cablin from different habitats.

ABSTRACT

Objective: To study difference of the immune response between oral administration and intraperitoneal injection of total polysaccharide of Sijunzi decoction (SJTP) in mice. Methods: The immunosuppressed mice model was established by intraperitoneal injection of cyclophosphamide (CY). The effects of SJTP on the weight of immune organs, hemolysin response and delayed type hypersensitivity (DTH) were observed, and the influences on normal mouse spleen lymphocytic proliferation of mice were also studied. Results:SJTP can remarkably counteract the immunosuppression induced by CY, decrease the weight loss immune organs, increase the hemolysin response and DTH,and promote the proliferation of lymphocytes. The effect of oral administration is superior to intrapertoneal injection. Conclusion:SJTP can improve the humoral and cellalar immunity of mice. The immune activities resulting from oral administration of SJTP have some special characteristics, and its possible mechanism is also discussed.
